The IBF world welterweight champ came in at 10 st 6lb 7oz, with his opponent virtually even at 10 st 6lb 5oz.
Brook will be fighting at his natural weight at the Sheffield City Hall after moving up two divisions to face big-hitter Gennady Golovkin.
The crowd cheered as Brook showed off an impressive physique before he squared up to highly-rated Spence on the stage.
Speaking to Sky Sports, Brook said: “I’m refuelling up, putting the fluids back in, rehydrating, putting the carbs back in.
“The part is now the fight. I’ve done the hard work by getting back down to welterweight – everyone thought I wouldn’t get there.
“Getting back down to the weight is always tough for me but has not been as terrible as usual this time because I’ve done it right.
